B and T lymphocyte attenuator (BTLA) is a member of the CD28 Ig superfamily of inhibitory co-receptors, along with CTLA-4 and PD-1. However, unlike CTLA-4 and PD-1, BTLA does not interact with B7-Ig family receptors. Rather, it is engaged by herpesvirus entry mediator (HVEM), a member of the TNF receptor superfamily, leading to an inhibited immune response. It also plays a role in immune tolerance. BTLA is expressed on most CD4 and CD8 T cells, B cells, macrophages, dentritic cells, and NK cell. This antibody recognizes a protein of 75 kDa, identified as Thrombomodulin. Thrombomodulin is a transmembrane glycoprotein with natural anticoagulant properties. It is normally expressed by a restricted number of cells, such as endothelial and mesothelial cells. In addition, synovial lining and syncytio-trophoblasts of placenta also express thrombomodulin. This protein is present in almost all of benign vascular tumors and majority of malignant vascular tumors (Kaposi's sarcoma, angiosarcoma, and epithelioid hemangioendothelioma). Hence, anti-thrombomodulin serves as a sensitive marker for lymphatic endothelial cells and their tumors. Defects in TNFRSF13C are the cause of immunodeficiency common variable type 4 (CVID4) ; also called antibody deficiency due to BAFFR defect. CVID4 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B cells is usually in the normal range, but can be low. This antibody recognizes a protein of 27-32 kDa, identified as Connexin 32. The connexin family of proteins forms hexameric complexes called connexons that facilitate movement of low molecular weight proteins between cells via gap junctions. Connexin proteins share a common topology of four transmembrane alpha-helicaldomains, two extracellular loops, a cytoplasmic loop and cytoplasmic N- and C-termini. Many of the key functional differences arise from specific amino-acid substitutions in the most highly conserved domains, the transmembrane and extracellular regions. This antibody reacts with E6 protein of human papilloma virus type 16 (HPV-16). Infection with specific types of HPV has been associated with an increased risk of developing cervical neoplasia. HPV types 6 and 11 have been associated with relatively benign diseases such as genital warts but types 16 and 18 are strongly associated with cervical, vaginal, and vulvar malignancies. Biotium Cytokeratin 10 (Suprabasal Epithelial Marker) (KRT10/1275), CF740 conjugate, 0.1mg/mL
This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Aldehyde dehydrogenase 1 family member A1 (ALDH1A1), also known as retinal dehydrogenase 1, belongs to the aldehyde dehydrogenase enzyme family, which is involved in the metabolism of alcohol. ALDH1A1 is predominantly expressed in the epithelium of testis, brain, eye, liver, kidney, as well as neural and hematopoietic stem cells. Reportedly, high ALDH1A1 expression is found in solitary fibrous tumor (SFT) and hemangiopericytoma (HPC), compared to meningiomas and synovial sarcomas. In combination with CD34, ALDH1A1 may be useful for the differentiation among SFT, HPC, meningioma, and synovial sarcoma. Human papilloma viruses (HPVs) can be classified as either high risk or low risk according to their association with cancer. HPV16 and HPV18 are the most common of the high risk group while HPV6 and HPV11 are among the low risk types. Approximately 90% of cervical cancers contain HPV DNA of the high risk types. Mutational analysis has shown that the E6 and E7 genes of the high risk HPVs are necessary and sufficient for HPV transforming function. The specific interactions of the E6 and E7 proteins with p53 and pRB, respectively, correlate with HPV high and low risk classifications. The high risk HPV E7 proteins bind to pRB with a higher affinity than do the low risk HPV proteins, and only the high risk HPV E6 proteins form detectable complexes with p53 in vitro. This antibody recognizes a protein of 30-33 kDa, which is identified as CD20. Its epitope is located in the cytoplasmic domain of CD20 and was, therefore, ascribed as CD20cy in the 5th Workshop. CD20 is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood and B cell localization in tissues. It reacts with the majority of B-cells present in peripheral blood and lymphoid tissues and their derived lymphomas. This antibody recognizes a protein of ~140 kDa, identified as N-Cadherin (NCAD), also known as CD325. NCAD is a member of the Cadherin superfamily, and consists of five extracellular repeats, a transmembrane domain and a cytoplasmic domain. CD325 deficient mice die at day 10 of gestation and embryos display major heart defects and malformed neural tubes and somites. Consistent with this, CD325 has been implicated in several aspects of cardiac development including the precardiac mesoderm, establishment of left-right symmetry and cardiac looping morphogenesis. Furthermore, CD325 is normally involved in inducing cell cycle arrest and its expression is frequently deregulated in cancer cells. Creatine kinases (CK) are a large family of isoenzymes that regulate levels of ATP in subcellular compartments, where they provide ATP at sites of fluctuating energy demand by the transfer of phosphates between creatine and adenine nucleotides. CKs provide the energy of phosphate hydrolysis necessary to drive the normal function of many cellular systems. In cells, the cytosolic CK enzymes consist of two subunits, which can be either B (brain type) or M (muscle type). There are three different isoenzymes: CKMM, CKBB and CKMB.
